Jean-Luc blow for Stormers
Du Plessis injured his ankle in his team's 52-24 bonus-point win over the Southern Kings on Saturday and Stormers head coach Robbie Fleck said it looked "serious".
Wing Kobus van Wyk is another concern for the Cape side after the 24-year-old left the field with a rib injury in the first half.
"Jean-Luc is the serious one – he's got an ankle injury which does not look good at the moment, but Kobus is hopefully okay," Fleck told reporters after the match.
"It looked like he [Van Wyk] might have damaged a rib.
"The initial feedback on Jean-Luc's injury is that it could be a syndesmosis. If that is the case, then it is usually four months out."
The good news for the Stormers is that they should have Robert du Preez back for the play-offs. Du Preez has recovered from a serious knee injury he picked up in March and has been playing with Western Province in the Currie Cup Qualifying tournament.
Brandon Thomson, who replaced Du Plessis in the Kings match, acquitted himself well on Saturday and is another option for the No.10 jersey.
"He [Du Preez] has played well for the Currie Cup side, while we were on tour.
"We will reassess the situation on Monday, but the pleasing aspect is that Rob is back to full fitness and [Brandon] Thomson got a bit of game time tonight, which is important," Fleck added.
Meanwhile, Fleck believes that they have ticked the right boxes in their last three matches against the Melbourne Rebels, Western Force and Kings.
The Cape side picked up a maximum 15 points from their last three matches, scoring 18 tries in total.
"We are starting to get that balance right now," said Fleck. "I think the subtleties in our game are starting to come through now and we are starting to get that variation right.
"That excites us and you need that variation against a quality outfit. I think we are putting teams under pressure in different parts of the game, which makes us a little bit less predictable and a little more dangerous.
"We scored 10 tries on tour and we scored eight [against the Kings] … The key for me was just the intensity the New Zealand sides were playing at and I guess that was a driving force from my side – to try and match that intensity and I think at stages we did get it right."
