Lambie was knocked out when Ireland flank CJ Stander clattered into him late while trying to charge down a kick in the first half. Lambie, who was motionless after the incident, had to be taken off with a stretcher
Stander received a red card for the incident and will appear before a disciplinary hearing in Cape Town on Sunday morning.
Coetzee said Lambie will miss the second Test against the Irish in Johannesburg, but he is hopeful the 25-year-old will be fit for the final Test in the three-match series in Port Elizabeth on June 25.
"The provisional diagnosis is that Pat was concussed and although he is ok, will not be considered for next weekend," Springbok coach Allister Coetzee said after the match.
Coetzee said a call on a possible replacement will be made later.
Garth April, who was part of the SA 'A' side that lost to the England Saxons on Friday, will likely join the squad as flyhalf cover.
April was part of Coetzee's 31-man squad before he was released to the SA 'A' side earlier in the week.
