Harumichi Tatekawa has been named as captain again.
Hammett made a number of changes for the trip to Loftus Versfeld.
Hitoshi Ono will start ahead of Fa'atiga Lemalu at lock, while at loosehead prop Keita Inagaki replaces Masataka Mikami in the No.1 jersey.
In the backline Hammett rotated Yuki Yatomi and Kaito Shigeno at scrumhalf, with the former starting in the No 9 jersey.
